چکیده

1. The plasma clearance of intravenously injected 'l25-labelled mitochondrial malate dehydrogenase (half-life 7 min) was not influenced by previous injection of suramin and/or leupeptin (inhibitors of intralysosomal proteolysis). 2. Pretreatment with both inhibitors considerably delayed degradation of endocytosed enzyme in liver, spleen, bone marrow and kidneys. 3. The tissue distribution of radioactivity was determined at 30min after injection, when only 3% of the dose was left in plasma. All injected radioactivity was still present in the carcass. The major part of the injected dose was found in liver (49%), spleen (5%), kidneys (13%) and bone, including marrow (11%). 4. Liver cells were isolated 15 min after injection of labelled enzyme. We found that Kupffer cells and parenchymal cells had endocytosed the enzyme at rates corresponding

Antagonistic Effects of Suramin Against the Venom of Snake, Echis Carinatus, on the Circulatory System of Developing Chicken Embryos

Background: The snake, Echis carinatus, one of the most venomous snakes in Asia, possesses a deadly hemotoxic venom. It has been reported that suramin, an anti-trypansomiasis drug, can inhibit the toxic effects of some snake venoms. This study was conducted to evaluate the possible antagonistic effects of suramin against the hemorrhagic activity of the venom from an Iranian snake, Echis carinat...

Short- and Long-Term Functional Alterations of the Skeletal Muscle Calcium Release Channel (Ryanodine Receptor) by Suramin: Apparent Dissociation of Single Channel Current Recording and [H]Ryanodine Binding

The present study demonstrates the following characteristic suramin actions on the purified skeletal muscle calcium release channel in single-channel current recordings and [H]ryanodine binding to HSR: 1) Suramin (0.3–0.9 mM) induced a concentration-dependent increase in the open probability (Po > 0.9) at 20 to 100 mM Ca and an almost fully open channel at 1 mM Ca (Po 5 0.95) with a marked shif...

Growth Factor Receptor Binding in HT-29 Cells Suramin Alters Phosphoinositide Synthesis and Inhibits

Initiation of cell growth frequently involves activation of growth factor receptor-coupled u rosine kinases and stimulation of the phosphoinositide second messenger system. The antitrypanosomal and antifiliarial drug suramin has been shown to exert antiproliferative activities by inhibition of growth factor receptor binding. We therefore investigated the effect of suramin on epidermal growth fa...

Suramin inhibits the early effects of PLA

Several phospholipase A 2 (PLA 2 ) neurotoxins from snake venoms can affect acetylcholine release at the neuromuscular junction. In isolated nerve-muscle preparations three distinct phases have been described for this phenomenon: An initial transient decrease in twitch tension; a second facilitatory phase during which twitch height is greater than control twitch height; and the last phase which...
